چکیده
Introduction: Isolation of fungi from tissue specimens using conventional methods is time consuming. However, in some cases, the histopathological examination (HPE) alone unable to provide a definite identity fungus. Alternatively, non-culture method, such as polymerase chain reaction (PCR) detecting internal transcribed spacer (ITS) rRNA genes fungi, promising diagnostic tool for rapid and accurate diagnosis mycoses. Methods: This work investigated utility panfungal PCR identifying agents mycoses 87 FFPE archive specimens. Deoxyribonucleic acid (DNA) extraction was performed on by QIAamp DNA Tissue Kit. The ITS2 region amplified ITS3/ITS4 primers. products were sequenced same primers compared NCBI nucleotide database species identification. Results: Fungal successfully 52 (59.8%) specimens, which only 23 (44.0%) consistent with clinical/HPE findings. identified Aspergillus spp., Candida Penicillium Cryptococcus neoformans, Talaromyces marneffei, Rhizopus oryzae. A few rare also identified, Diaporthe longicolla fungus-like oomycete Pythium insidiosum that are commonly associated plant pathogens. Conclusion: Although able offer genus/species identification, utilising this method paraffinised must be evaluated considering many factors will reduce its sensitivity specificity. Therefore, it important correlate results clinical HPE findings obtain correct adequate treatment

Toward deciphering proteomes of formalin-fixed paraffin-embedded (FFPE) tissues
Formalin-fixed paraffin-embedded (FFPE) tissue specimens comprise a potentially valuable resource for both prospective and retrospective biomarker discovery. Unlocking the proteomic profile of clinicopathological FFPE tissues is a critically essential step for annotating clinical findings and predicting biomarkers for ultimate disease prognosis and therapeutic follow-up.
متن کاملMolecular Identification of Mycobacterium Tuberculosis Complex in Formalin-Fixed, Paraffin-Embedded Tissue Blocks of Extra Pulmonary Speciemens using Genomics Extraction
Background: Tuberculosis has been detected in some extra pulmonary ecological niches. Although extra pulmonary tuberculosis (EPTB) is less frequent than Pulmonary Tuberculosis (PTB), its incidence has increased worldwide. The aim of this study was to investigate the presence of EPTB and MDR-EXPT in Formalin-fixed, paraffin-embedded tissue blocks among different samples in Kerma...
متن کاملTissue lithography: Microscale dewaxing to enable retrospective studies on formalin-fixed paraffin-embedded (FFPE) tissue sections
We present a new concept, termed tissue lithography (TL), and its implementation which enables retrospective studies on formalin-fixed paraffin-embedded tissue sections. Tissue lithography uses a microfluidic probe to remove microscale areas of the paraffin layer on formalin-fixed paraffin-embedded biopsy samples. Current practices in sample utilization for research and diagnostics require comp...
متن کاملTranscriptome Profiling of Archived Sectioned Formalin-Fixed Paraffin-Embedded (AS-FFPE) Tissue for Disease Classification
BACKGROUND Archived tissues from previously completed prospective trials represent invaluable resource for biomarker development. However, such specimens are often stored as sections on glass slides, in which RNA is severely degraded due to prolonged air exposure. We evaluated whether a proportion of archived sectioned formalin-fixed paraffin-embedded (AS-FFPE) tissues yield transcriptome profi...
